This variant is present in population databases (rs565591812, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with MYPN-related conditions. ClinVar contains an entry for this variant (Variation ID: 569410). The p.R1044W variant (also known as c.3130C>T), located in coding exon 14 of the MYPN gene, results from a C to T substitution at nucleotide position 3130. The arginine at codon 1044 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
