The c.116C>G (p.A39G) alteration is located in exon 1 (coding exon 1) of the KIF1BP gene. This alteration results from a C to G substitution at nucleotide position 116, causing the alanine (A) at amino acid position 39 to be replaced by a glycine (G). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

The alanine residue is highly conserved and there is a small physicochemical difference between alanine and glycine. This variant is present in population databases (rs372223602, ExAC 0.04%). This variant has not been reported in the literature in individuals affected with KIF1BP-related conditions. ClinVar contains an entry for this variant (Variation ID: 878713). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
