Genes affected
CDH23 (HGNC:13733): (cadherin related 23) This gene is a member of the cadherin superfamily, whose genes encode calcium dependent cell-cell adhesion glycoproteins. The encoded protein is thought to be involved in stereocilia organization and hair bundle formation. The gene is located in a region containing the human deafness loci DFNB12 and USH1D. Usher syndrome 1D and nonsyndromic autosomal recessive deafness DFNB12 are caused by allelic mutations of this cadherin-like gene. Upregulation of this gene may also be associated with breast cancer. Alternative splice variants encoding different isoforms have been described. [provided by RefSeq, May 2013]
C10orf105 (HGNC:20304): (chromosome 10 open reading frame 105) Predicted to be integral component of membrane. [provided by Alliance of Genome Resources, Apr 2022]

Uncertain significance, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpMar 24, 2022This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 1076 of the CDH23 protein (p.Gly1076Arg). This variant is present in population databases (no rsID available, gnomAD 0.007%). This missense change has been observed in individual(s) with deafness (Invitae). ClinVar contains an entry for this variant (Variation ID: 1013285). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Not Available"; Align-GVGD: "Class C65"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
Usher syndrome type 1D Uncertain:1
Uncertain significance, criteria provided, single submitterclinical testingPittsburgh Clinical Genomics Laboratory, University of Pittsburgh Medical CenterFeb 09, 2024This sequence variant is a single nucleotide substitution (G>C) at position 3226 of the coding sequence of the CDH23 gene that results in a glycine to arginine amino acid change at residue 1076 of the cadherin related 23 protein. This residue falls in the extracellular cadherin 10 domain of the protein (UniProt). This is a previously reported variant (ClinVar 1013285) that has not been observed in the literature in individuals affected by CDH23-related disease, to our knowledge. This variant is present in 3 of 1613362 alleles (0.0001859%) in the gnomAD v4.0.0 population dataset. Multiple bioinformatic tools predict that this amino acid change would be neutral, and the Gly1076 residue at this position is highly conserved across the vertebrate species examined. Studies examining the functional consequence of this variant have not been performed, to our knowledge. At this time, there is insufficient evidence to determine if this variant is pathogenic or benign. Therefore, we consider this a variant of uncertain significance. ACMG Criteria: BP4, PM2, PM3 -
