R570X was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The R570X variant is predicted to cause loss of normal protein function either by protein truncation or nonsense-mediated mRNA decay. However, other nonsense mutations in the VCL gene have not been reported in association with cardiomyopathy.With the clinical and molecular information available at this time, we cannot definitively determine if R570X is a disease-causing mutation or a rare benign variant.
