VCL (HGNC:12665): (vinculin) Vinculin is a cytoskeletal protein associated with cell-cell and cell-matrix junctions, where it is thought to function as one of several interacting proteins involved in anchoring F-actin to the membrane. Defects in VCL are the cause of cardiomyopathy dilated type 1W. Dilated cardiomyopathy is a disorder characterized by ventricular dilation and impaired systolic function, resulting in congestive heart failure and arrhythmia. Multiple alternatively spliced transcript variants have been found for this gene, but the biological validity of some variants has not been determined. [provided by RefSeq, Jul 2008]

ClinVar contains an entry for this variant (Variation ID: 202161). This sequence change creates a premature translational stop signal at codon 570 (p.Arg570*) of the VCL gene. It is expected to result in an absent or disrupted protein product. -

R570X was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The R570X variant is predicted to cause loss of normal protein function either by protein truncation or nonsense-mediated mRNA decay. However, other nonsense mutations in the VCL gene have not been reported in association with cardiomyopathy.With the clinical and molecular information available at this time, we cannot definitively determine if R570X is a disease-causing mutation or a rare benign variant. The variant is found in DCM-CRDM panel(s). -
