Genes affected
ADK (HGNC:257): (adenosine kinase) This gene an enzyme which catalyzes the transfer of the gamma-phosphate from ATP to adenosine, thereby serving as a regulator of concentrations of both extracellular adenosine and intracellular adenine nucleotides. Adenosine has widespread effects on the cardiovascular, nervous, respiratory, and immune systems and inhibitors of the enzyme could play an important pharmacological role in increasing intravascular adenosine concentrations and acting as anti-inflammatory agents. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jan 2011]

Adenosine kinase deficiency Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Pittsburgh Clinical Genomics Laboratory, University of Pittsburgh Medical Center | Jan 22, 2024 | This sequence variant is a single nucleotide substitution (T>C) at position 79 of the coding sequence of the ADK gene that results in a phenylalanine to leucine amino acid change at residue 27 of the adenosine kinase protein. Residue 27 falls in the kinase domain of the protein (UniProt). This variant is absent from ClinVar and has not been observed in individuals affected by an ADK-related disorder in the published literature, to our knowledge. This variant is present in 32 of 1610670 alleles (0.0020%) in the gnomAD v4.0.0 population dataset. Bioinformatic tools are inconclusive if this amino acid change will be damaging or tolerated, and the Phe27 residue at this position is highly conserved across the vertebrate species examined. Studies examining the functional consequence of this variant have not been published, to our knowledge. At this time, there is insufficient evidence to determine if this variant is pathogenic or benign. Therefore, we consider this a variant of uncertain significance. ACMG Criteria: PM2 - |
