The proline at codon 281 is replaced by alanine, an amino acid with highly similar properties. In one study, this variant was reported in 0/60,466 breast cancer cases and in 1/53,461 controls (Dorling et al. N Engl J Med. 2021 02;384:428-439). This alteration was also identified in a cohort of pancreatic cancer patients undergoing multigene panel testing (Young EL et al. BMC Cancer, 2018 Jun;18:697). This variant demonstrated wild-type like intracellular protein abundance on one multiplex functional assay, as well as wild-type like lipid phosphatase activity in a massively parallel functional assay using a humanized yeast model (Matreyek KA et al. Nat Genet, 2018 06;50:874-882; Mighell TL et al. Am J Hum Genet, 2018 05;102:943-955). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
