A variant of uncertain significance has been identified in the TCTN3 gene. The L592V variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. The L592V variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The L592V variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is not conserved, and in silico analysis predicts this variant likely does not alter the protein structure/function. Based on the currently available information, it is unclear whether this variant is a pathogenic variant or a rare benign variant. -

A heterozygous missense variant was identified, NM_015631.5(TCTN3):c.1774C>G in exon 14 of 14 of the TCTN3 gene. This substitution is predicted to create a minor amino acid change from leucine to valine at position 592 of the protein, NP_056446.4(TCTN3):p.(Leu592Val). The leucine at this position has low conservation (100 vertebrates, UCSC), and it is not situated in a known functional domain. In silico software predicts this variant to be benign (Polyphen, SIFT, CADD, Mutation Taster). The variant is present in the gnomAD population database at a frequency of 0.0046% (13 heterozygotes, 0 homozygotes).
