The c.1696G>A (p.G566S) alteration is located in exon 12 (coding exon 12) of the HPSE2 gene. This alteration results from a G to A substitution at nucleotide position 1696, causing the glycine (G) at amino acid position 566 to be replaced by a serine (S). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with HPSE2-related disease. This variant is present in population databases (rs577966759, ExAC 0.002%).
