This variant has not, to our knowledge, been published in the literature as pathogenic or benign. ATM Val27Ile was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, suggesting it is not a common benign variant in these populations. Since Valine and Isoleucine share similar properties, this is considered a conservative amino acid substitution. ATM Val27Ile occurs at a position where amino acids with properties similar to Valine are tolerated across species and is not located in a known functional domain (UniProt). In silico analyses predict that this variant is unlikely to alter protein structure or function. Based on currently available evidence, it is unclear whether ATM Val27Ile is a pathogenic or benign variant. We consider it to be a variant of uncertain significance. - |

The valine at codon 27 is replaced by isoleucine, an amino acid with highly similar properties. This alteration was identified in a Spanish breast and/or ovarian cancer family (Tavera-Tapia A et al. Breast Cancer Res Treat, 2017 02;161:597-604). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
