The serine at codon 49 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This variant was identified as germline in a patient with a pancreatic neuroendocrine tumor that demonstrated loss of heterozygosity (LOH) at this position (Lawrence B et al. NPJ Genom Med, 2018 Jul;3:18). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
