This sequence change replaces lysine with arginine at codon 106 of the ATM protein (p.Lys106Arg). The lysine residue is weakly conserved and there is a small physicochemical difference between lysine and arginine. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a ATM-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. A lysine is also found at this position in multiple mammalian species indicating this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies. In summary, this is a novel missense change with uncertain impact mRNA splicing and protein function. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of nucleotide changes on mRNA splicing suggest that this variant may alter mRNA splicing, but this prediction has not been confirmed by published transcriptional studies. -
