This missense variant replaces leucine with proline at codon 651 of the ATM protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with breast cancer as well as in unaffected controls (PMID: 19404735). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.L651P variant (also known as c.1952T>C), located in coding exon 12 of the ATM gene, results from a T to C substitution at nucleotide position 1952. The leucine at codon 651 is replaced by proline, an amino acid with similar properties. This variant was identified in 1/122 early-onset breast cancer cases who also had family history of breast and/or ovarian cancer as well as hematological malignancy and in 4/186 controls (Paglia LL et al. Breast Cancer Res. Treat., 2010 Jan;119:443-52). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
