This variant is denoted ATM c.2192A>T at the cDNA level, p.Tyr731Phe (Y731F) at the protein level, and results in the change of a Tyrosine to a Phenylalanine (TAC>TTC). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. ATM Tyr731Phe was not observed in large population cohorts (Lek 2016). Since Tyrosine and Phenylalanine differ in polarity, charge, size or other properties, this is considered a non-conservative amino acid substitution. ATM Tyr731Phe is not located in a known functional domain. In-silico analyses, including protein predictors and evolutionary conservation, support that this variant does not alter protein structure or function. The p.Y731F variant (also known as c.2192A>T), located in coding exon 13 of the ATM gene, results from an A to T substitution at nucleotide position 2192. The tyrosine at codon 731 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
