However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 1.6e-05 in 246118 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.4482T>C in individuals affected with Breast Cancer and no experimental evidence demonstrating its impact on protein function have been reported. Co-occurrences with other pathogenic variant(s) have been reported (BRCA2 c.5290_5291delTC, p.Ser1764fsX3), providing supporting evidence for a benign role. Three ClinVar submissions from clinical diagnostic laboratories (evaluation after 2014) cite the variant as likely benign.
