This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with ATM-related conditions. ClinVar contains an entry for this variant (Variation ID: 1389273). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. This variant disrupts the p.Thr1743 amino acid residue in ATM. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 9463314, 17968022, 19147735, 19431188, 19781682, 21792198, 21933854, 27978560, 30303537, 31921190). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. The p.T1743S variant (also known as c.5227A>T), located in coding exon 34 of the ATM gene, results from an A to T substitution at nucleotide position 5227. The threonine at codon 1743 is replaced by serine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
