Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2.4e-05 in 250924 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.5470C>T has been reported in the literature in an individual affected with breast cancer (Decker_2017). In contrast, the variant was not found in 5560 individuals affected with prostate cancer from the PRACTICAL Consortium study groups cohort, but was in 1 of 3353 controls of European ancestry (Karlsson_2021). These reports do not provide unequivocal conclusions about association of the variant with breast or prostate cancers. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 28779002, 33436325). Seven submitters have cited clinical-significance assessments for this variant to ClinVar after 2014 and all classified the variant as uncertain significance.
