ATM (HGNC:795): (ATM serine/threonine kinase) The protein encoded by this gene belongs to the PI3/PI4-kinase family. This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. Mutations in this gene are associated with ataxia telangiectasia, an autosomal recessive disorder. [provided by RefSeq, Aug 2010]

The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.5470C>T has been reported in the literature in an individual affected with breast cancer (Decker_2017). In contrast, the variant was not found in 5560 individuals affected with prostate cancer from the PRACTICAL Consortium study groups cohort, but was in 1 of 3353 controls of European ancestry (Karlsson_2021). These reports do not provide unequivocal conclusions about association of the variant with breast or prostate cancers. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 28779002, 33436325). Seven submitters have cited clinical-significance assessments for this variant to ClinVar after 2014 and all classified the variant as uncertain significance.
