The p.R2443L variant (also known as c.7328G>T), located in coding exon 49 of the ATM gene, results from a G to T substitution at nucleotide position 7328. The arginine at codon 2443 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
This missense variant replaces arginine with leucine at codon 2443 of the ATM protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with breast cancer who also carried a pathogenic co-variant in BRCA2 (PMID: 32868316). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This sequence change replaces arginine, which is basic and polar, with leucine, which is neutral and non-polar, at codon 2443 of the ATM protein (p.Arg2443Leu).
