Four of five in-silico tools predict a benign effect of the variant on protein function. Several computational tools predict a significant impact on normal splicing: Two predict the variant creates a 5' donor site. One predict the variant strengthens a cryptic 5' donor site. Three predict the variant strengthens a 3' acceptor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 3e-05 in 1612730 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in ATM causing Breast Cancer (3e-05 vs 0.001), allowing no conclusion about variant significance. Additionally, this variant was carried by 2 individuals in the FLOSSIES database (individuals >70 years of age with no personal history of cancer). To our knowledge, no occurrence of c.7516A>G in individuals affected with ATM-related conditions and no experimental evidence demonstrating its impact on protein function have been reported.
