The phenylalanine at codon 2686 is replaced by leucine, an amino acid with highly similar properties. This alteration was identified in an individual with early onset cerebellar ataxia. This individual also carried ATM c.8151+1G>A however phase was not described (Shakya S et al. Clin Genet, 2019 12;96:566-574). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
