Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Jan 19, 2024 | This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 40 of the SDHD protein (p.Ile40Val). This variant is present in population databases (rs146276662, gnomAD 0.004%). This missense change has been observed in individual(s) with a personal or family history of paraganglioma (PMID: 19351833, 29386252). ClinVar contains an entry for this variant (Variation ID: 465221). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on SDHD protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. - |
not provided Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | GeneDx | May 19, 2022 | Not observed at significant frequency in large population cohorts (gnomAD); In silico analysis, which includes protein predictors and evolutionary conservation, supports that this variant does not alter protein structure/function; Observed in an individual with a head/neck paraganglioma (Neumann 2009); This variant is associated with the following publications: (PMID: 19351833, 29386252, 28748451) - |
Hereditary cancer-predisposing syndrome Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Jul 17, 2024 | The p.I40V variant (also known as c.118A>G), located in coding exon 2 of the SDHD gene, results from an A to G substitution at nucleotide position 118. The isoleucine at codon 40 is replaced by valine, an amino acid with highly similar properties. In a study of 598 unrelated probands diagnosed with head and neck paraganglioma, this alteration was detected in 1 individual (Neumann HP et al. Cancer Res., 2009 Apr;69:3650-6). This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. - |
