Uncertain significance, criteria provided, single submitter | clinical testing | GeneDx | May 25, 2023 | In silico analysis supports that this missense variant does not alter protein structure/function; Has not been previously published as pathogenic or benign to our knowledge; This variant is associated with the following publications: (PMID: 14985401) - |
not specified Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Genetic Services Laboratory, University of Chicago | Jun 09, 2021 | DNA sequence analysis of the SDHD gene demonstrated a sequence change, c.269C>T, in exon 3 that results in an amino acid change, p.Ala90Val. This sequence change has been described gnomAD with a frequency of 0.091% in the South Asian sub-population (dbSNP rs200255724). The p.Ala90Val change affects a poorly conserved amino acid residue located in a domain of the SDHD protein that is known to be functional. The p.Ala90Val substitution appears to be tolerated using several in-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL). This sequence change does not appear to have been previously described in patients with SDHD-related disorders. Due to the lack of sufficient evidences, the clinical significance of the p.Ala90Val change remains unknown at this time. - |

Hereditary cancer-predisposing syndrome Benign:1
Likely benign, criteria provided, single submitter | clinical testing | Ambry Genetics | Oct 04, 2021 | This alteration is classified as likely benign based on a combination of the following: seen in unaffected individuals, population frequency, intact protein function, lack of segregation with disease, co-occurrence, RNA analysis, in silico models, amino acid conservation, lack of disease association in case-control studies, and/or the mechanism of disease or impacted region is inconsistent with a known cause of pathogenicity. - |
