The p.D541V variant (also known as c.1622A>T), located in coding exon 11 of the CBL gene, results from an A to T substitution at nucleotide position 1622. The aspartic acid at codon 541 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -

This sequence change replaces aspartic acid, which is acidic and polar, with valine, which is neutral and non-polar, at codon 541 of the CBL protein (p.Asp541Val). This variant is present in population databases (rs147285276, gnomAD 0.005%). This variant has not been reported in the literature in individuals affected with CBL-related conditions.
