The p.E699D variant (also known as c.2097G>C), located in coding exon 13 of the CBL gene, results from a G to C substitution at nucleotide position 2097. The glutamic acid at codon 699 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.

This sequence change replaces glutamic acid, which is acidic and polar, with aspartic acid, which is acidic and polar, at codon 699 of the CBL protein (p.Glu699Asp).
