CBL-related disorder Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | PreventionGenetics, part of Exact Sciences | Jul 27, 2023 | The CBL c.2350G>A variant is predicted to result in the amino acid substitution p.Val784Met. This variant was reported in an individual with clinical suspicion of RASopathy (Lepri et al. 2014. PubMed ID: 24451042). This variant is reported in 0.0062% of alleles in individuals of European (Non-Finnish) descent in gnomAD (http://gnomad.broadinstitute.org/variant/11-119169166-G-A). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. - |
not provided Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | GeneDx | Jun 08, 2015 | The V784M missense change in the CBL gene has been reported previously in association with Noonan spectrum disorder (Lepri et al., 2014). It was not observed at any significant frequency in approximately 6,400 individuals of European and African American ancestry by the NHLBI Exome Sequencing Project. V784M is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is conserved in mammals. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. Based on the currently available information, it is unclear whether this variant is a pathogenic mutation or a rare benign variant. - |
RASopathy Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Dec 28, 2023 | This sequence change replaces valine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 784 of the CBL protein (p.Val784Met). This variant is present in population databases (rs140725852, gnomAD 0.006%). This missense change has been observed in individual(s) with Noonan syndrome (PMID: 24451042). ClinVar contains an entry for this variant (Variation ID: 40422). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CBL protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. - |
