TECTA (HGNC:11720): (tectorin alpha) The tectorial membrane is an extracellular matrix of the inner ear that contacts the stereocilia bundles of specialized sensory hair cells. Sound induces movement of these hair cells relative to the tectorial membrane, deflects the stereocilia, and leads to fluctuations in hair-cell membrane potential, transducing sound into electrical signals. Alpha-tectorin is one of the major noncollagenous components of the tectorial membrane. Mutations in the TECTA gene have been shown to be responsible for autosomal dominant nonsyndromic hearing impairment and a recessive form of sensorineural pre-lingual non-syndromic deafness. This variant has been observed in twelve affected individuals and one unaffected individual of a large family with deafness (PMID: 22718023); Missense variant consistently predicted to be damaging by in silico tool or highly conserved with a major amino acid change. Additional information: Variant is predicted to result in a missense amino acid change from isoleucine to threonine; This variant is heterozygous; This gene is associated with both recessive and dominant disease. Generally, biallelic loss of function variants cause recessive deafness, while missense variants cause dominant deafness (PMID: 9949200, 20947814, 28946916) - Previous reports of pathogenicity for this variant are conflicting. This variant has been classified as a VUS by a clinical laboratory in ClinVar, and has been reported in the literature in two families with deafness (PMID: 22718023); No published functional evidence has been identified for this variant; No comparable missense variants have previous evidence for pathogenicity; Variant is located in the Zona pellucida-like domain (DECIPHER); Loss of function is a known mechanism of disease in this gene and is associated with autosomal recessive deafness 21 (MIM#603629).
