Genes affected
PTH (HGNC:9606): (parathyroid hormone) This gene encodes a member of the parathyroid family of proteins. The encoded preproprotein is proteolytically processed to generate a protein that binds to the parathyroid hormone/parathyroid hormone-related peptide receptor and regulates blood calcium and phosphate levels. Excess production of the encoded protein, known as hyperparathyroidism, can result in hypercalcemia and kidney stones. On the other hand, defective processing of the encoded protein may lead to hypoparathyroidism, which can result in hypocalcemia and numbness. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Oct 2015]

Uncertain significance, criteria provided, single submitterclinical testingAmbry GeneticsJun 03, 2024The c.230C>A (p.A77D) alteration is located in exon 3 (coding exon 2) of the PTH gene. This alteration results from a C to A substitution at nucleotide position 230, causing the alanine (A) at amino acid position 77 to be replaced by an aspartic acid (D). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

Uncertain significance, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpJan 30, 2023This sequence change replaces alanine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 77 of the PTH protein (p.Ala77Asp). This variant is present in population databases (rs201237944, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with PTH-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available". The aspartic acid amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
