This sequence change replaces arginine with leucine at codon 1075 of the OTOG protein (p.Arg1075Leu). The arginine residue is moderately conserved and there is a moderate physicochemical difference between arginine and leucine. While this variant is present in population databases (rs11024333), the frequency information is unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has not been reported in the literature in individuals affected with OTOG-related conditions.
