It is the rate-limiting enzyme in the synthesis of catecholamines, hence plays a key role in the physiology of adrenergic neurons. Variant summary: TH c.832G>A (p.Ala278Thr) results in a non-conservative amino acid change located in the Aromatic amino acid hydroxylase, C-terminal domain (IPR019774) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.832G>A has been reported in the literature in an individual affected with Segawa Syndrome, Autosomal Recessive (Wang_2022). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. This variant is also known as c.739G>A(p.A247T). The following publication have been ascertained in the context of this evaluation (PMID: 36568392). This sequence change replaces alanine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 278 of the TH protein (p.Ala278Thr).
