Genes affected
ANO5 (HGNC:27337): (anoctamin 5) This gene encodes a member of the anoctamin family of transmembrane proteins. The encoded protein is likely a calcium activated chloride channel. Mutations in this gene have been associated with gnathodiaphyseal dysplasia. Alternatively spliced transcript variants have been described. [provided by RefSeq, Nov 2009]

For these reasons, this variant has been classified as Pathogenic. This variant disrupts the p.Trp655 amino acid residue in ANO5. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 30919934; Invitae). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. This sequence change replaces tryptophan, which is neutral and slightly polar, with cysteine, which is neutral and slightly polar, at codon 655 of the ANO5 protein (p.Trp655Cys). This variant is present in population databases (rs760137559, gnomAD 0.006%). This missense change has been observed in individuals with autosomal recessive ANO5-related conditions (PMID: 22499103, 27447704). ClinVar contains an entry for this variant (Variation ID: 523571). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt ANO5 protein function.
