Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Nov 09, 2021 | The p.A283V variant (also known as c.848C>T), located in coding exon 6 of the KCNQ1 gene, results from a C to T substitution at nucleotide position 848. The alanine at codon 283 is replaced by valine, an amino acid with similar properties, and is located in the transmembrane-spanning region of the protein. This alteration has been reported in a sudden unexplained death in infancy cohort; however, clinical details were limited and additional alterations in genes associated with arrhythmias were also identified (Oshima Y et al. J Hum Genet, 2017 Nov;62:989-995). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
