This variant is present in population databases (rs730880569, gnomAD 0.002%). This missense change has been observed in individual(s) with hypertrophic cardiomyopathy (PMID: 33782553). The p.A848G variant (also known as c.2543C>G), located in coding exon 25 of the MYBPC3 gene, results from a C to G substitution at nucleotide position 2543. The alanine at codon 848 is replaced by glycine, an amino acid with similar properties. This alteration has been reported in a hypertrophic cardiomyopathy (HCM) cohort (Helms AS et al. Circ Genom Precis Med, 2020 Oct;13:396-405). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
