Myosin-binding protein C is a myosin-associated protein found in the cross-bridge-bearing zone (C region) of A bands in striated muscle. MYBPC3 is expressed exclusively in heart muscle and is a key regulator of cardiac contraction. Mutations in this gene are a frequent cause of familial hypertrophic cardiomyopathy. The variant is predicted to lead to either an abnormal message that is subject to nonsense-mediated mRNA decay, or to an abnormal protein product if the message is used for protein translation. Other splice site variants, including a pathogenic variant in the same splice acceptor site (c.906-1 G>C), have been reported in HGMD in association with hypertrophic cardiomyopathy (Stenson et al., 2014). Furthermore, the c.906-1 G>A variant was not observed in approximately 6,100 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. - |

Disruption of this splice site has been observed in individual(s) with clinical features of MYBPC3-related conditions (PMID: 18337725). ClinVar contains an entry for this variant (Variation ID: 279971). Studies have shown that disruption of this splice site results in skipping of 2 nucleotides of exon 10 and introduces a premature termination codon (PMID: 18337725). The resulting mRNA is expected to undergo nonsense-mediated decay. For these reasons, this variant has been classified as Pathogenic. - |
