Variant summary: MEN1 c.1022G>A (p.Trp341X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. Truncations downstream of this position have been classified as pathogenic by our laboratory. The variant was absent in 251458 control chromosomes. c.1022G>A has been reported in the literature in individuals affected with Multiple Endocrine Neoplasia Type 1 (example, Bassett_1998, Marini_2018). These data indicate that the variant is likely to be associated with disease. Two clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. All laboratories classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -

The p.W341* pathogenic mutation (also known as c.1022G>A), located in coding exon 6 of the MEN1 gene, results from a G to A substitution at nucleotide position 1022. This changes the amino acid from a tryptophan to a stop codon within coding exon 6. This alteration has been previously reported in individuals with multiple endocrine neoplasia type 1 (MEN1) (Bassett JH et al. Am J Hum Genet. 1998 Feb;62(2):232-44; Cebrian A et al. Eur J Hum Genet. 1999 Jul;7(5):585-9). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
