This pathogenic mutation has been identified in multiple families meeting criteria for MEN1 (Crépin M et al. Electrophoresis, 2003 Jan;24:26-33; Frank-Raue K et al. J. Clin. Endocrinol. Metab. 2005 Jul;90:4063-7). In addition to the clinical data presented in the literature, alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is classified as a disease-causing mutation. -
