Variant summary: MEN1 c.655-5C>G alters a nucleotide located at a position not widely known to affect splicing. Several computational tools predict a significant impact on normal splicing: Two predict the variant weakens a 3' acceptor site. Four predict the variant creates a 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 250756 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.655-5C>G in individuals affected with Multiple Endocrine Neoplasia Type 1 and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 241821). Based on the evidence outlined above, the variant was classified as uncertain significance. -
