This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 115 of the MEN1 protein (p.Arg115His). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with hereditary breast and/or ovarian cancer (PMID: 31159747). ClinVar contains an entry for this variant (Variation ID: 428047). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt MEN1 protein function with a positive predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.R115H variant (also known as c.344G>A) is located in coding exon 1 of the MEN1 gene. This alteration results from a G to A substitution at nucleotide position 344. The arginine at codon 115 is replaced by histidine, an amino acid with some highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of p.R115H remains unclear. -
