Variant summary
Our verdict is Uncertain significance. The variant received 2 ACMG points: 2P and 0B. PM2
The NM_016938.5(EFEMP2):c.186T>A(p.Pro62Pro) variant causes a synonymous change involving the alteration of a non-conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★). Synonymous variant affecting the same amino acid position (i.e. P62P) has been classified as Likely benign.

The c.186T>A variant (also known as p.P62P), located in coding exon 3 of the EFEMP2 gene, results from a T to A substitution at nucleotide position 186. This nucleotide substitution does not change the proline at codon 62. This nucleotide position is poorly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may result in the creation or strengthening of a novel splice acceptor site. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
