The V49A variant has notbeen published as pathogenic or been reported as benign to our knowledge. In addition, it is notobserved at a significant frequency in large population cohorts (Lek et al., 2016; 1000 GenomesConsortium et al., 2015; Exome Variant Server). Although the V49A variant is a conservative aminoacid substitution, which is not likely to impact secondary protein structure as these residues sharesimilar properties, this substitution does occur at a position that is conserved across species. Moreover,in silico analysis predicts this variant is probably damaging to the protein structure/function.Nonetheless, this variant lacks observation in a significant number of affected individuals, segregationdata, and functional evidence, which would further clarify its pathogenicity. -

This variant has not been reported in the literature in individuals affected with ILK-related conditions. This variant is present in population databases (rs201499797, gnomAD 0.0009%). This sequence change replaces valine, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 49 of the ILK protein (p.Val49Ala). ClinVar contains an entry for this variant (Variation ID: 432620). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
