This variant was reported in the compound heterozygous state with a second NADSYN1 variant an individual with cardiac and vertebral defects (overlapping with VACTERL) (Kortbawi et al 2022. PubMed ID: 35491967). This variant is reported in 0.0087% of alleles in individuals of Latino descent in gnomAD. Although we suspect this variant is pathogenic, at this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
