Autosomal recessive nonsyndromic hearing loss 2 Pathogenic:1
MYO7A c.5101C>T, p.R1701* is homozygous in a Palestinian child with severe to profound pre-lingual hearing loss (Abu Rayyan 2020). The variant is absent from 1300 Palestinian controls and absent from gnomAD v2.1.1. -

Usher syndrome type 1 Pathogenic:1
The stop gained variant c.5101C>T p.Arg1701Ter in the MYO7A gene has been reported in an individual in a homozygous state affected with Usher syndrome Khalaileh et al., 2018; Le Quesne Stabej et al., 2012. The variant has 0.001% allele frequency in gnomAD Exomes and is novel not in any individuals in 1000 Genomes. This variant has been reported to the ClinVar database as Likely Pathogenic/ Pathogenic. This variant is predicted to cause a loss of normal protein function through protein truncation. Loss of function variants has been previously reported to be disease causing. For these reasons, this variant has been classified as Pathogenic. -
not provided Pathogenic:1
This sequence change creates a premature translational stop signal (p.Arg1701*) in the MYO7A gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MYO7A are known to be pathogenic (PMID: 8900236, 25404053). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the gnomAD database. This premature translational stop signal has been observed in individual(s) with Usher syndrome (PMID: 16400615, 22135276, 29490346, 31266775). ClinVar contains an entry for this variant (Variation ID: 43270). For these reasons, this variant has been classified as Pathogenic. -

Rare genetic deafness Pathogenic:1
The Arg1701X variant in MYO7A has been reported as a compound heterozygous varia nt in one proband with Usher syndrome (Gerber 2006). In addition, this variant h as been identified by our laboratory as a homozygous variant in two siblings wit h clinical features of Usher syndrome. The Arg1701X variant leads to a premature stop codon at position 1701, which is predicted to lead to a truncated or absen t protein. In summary, this variant meets our criteria to be classified as patho genic. -
