The isoleucine at codon 1277 is replaced by methionine, an amino acid with highly similar properties. This alteration was detected in the heterozygous state in one reported patient with Charcot-Marie-Tooth disease; however, a second SBF2 alteration was not identified in this patient, and the alteration was also present in one control (Høyer H et al. Biomed Res Int, 2014 Jun;2014:210401). This amino acid position is highly conserved through mammals but not in all available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
