This sequence change replaces valine, which is neutral and non-polar, with isoleucine, which is neutral and non-polar, at codon 620 of the TRPV4 protein (p.Val620Ile). This variant is present in population databases (rs121912633, gnomAD 0.0009%). This missense change has been observed in individual(s) with autosomal dominant brachyolmia and hereditary motor and sensory neuropathy 2 (PMID: 18587396, 20460441). In at least one individual the variant was observed to be de novo. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 4993). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant.
