This sequence change replaces proline, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 35 of the CDKN1B protein (p.Pro35Leu). This variant is present in population databases (rs375297371, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with CDKN1B-related conditions. ClinVar contains an entry for this variant (Variation ID: 404279). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change does not substantially affect CDKN1B function (PMID: 28425505). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

Hereditary cancer-predisposing syndrome Uncertain:1
The p.P35L variant (also known as c.104C>T), located in coding exon 1 of the CDKN1B gene, results from a C to T substitution at nucleotide position 104. The proline at codon 35 is replaced by leucine, an amino acid with similar properties. One study found that this alteration does not significantly perturb binding with the VHL protein (Minervini G et al. Sci Rep, 2017 Apr;7:46562). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
