The p.P2147T variant (also known as c.6439C>A), located in coding exon 46 of the POLE gene, results from a C to A substitution at nucleotide position 6439. The proline at codon 2147 is replaced by threonine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
