This sequence change replaces glutamine, which is neutral and polar, with glutamic acid, which is acidic and polar, at codon 2121 of the POLE protein (p.Gln2121Glu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with POLE-related conditions. ClinVar contains an entry for this variant (Variation ID: 540721). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt POLE protein function with a negative predictive value of 80%. This alteration results from a C to G substitution at nucleotide position 6361, causing the glutamine (Q) at amino acid position 2121 to be replaced by a glutamic acid (E).
