This sequence change replaces glycine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 1924 of the POLE protein (p.Gly1924Arg). This variant is present in population databases (rs371862779, gnomAD 0.005%). This variant has not been reported in the literature in individuals affected with POLE-related conditions. ClinVar contains an entry for this variant (Variation ID: 246404). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt POLE protein function with a positive predictive value of 80%. The p.G1924R variant (also known as c.5770G>A), located in coding exon 42 of the POLE gene, results from a G to A substitution at nucleotide position 5770. The glycine at codon 1924 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
