In silico analysis supports that this missense variant does not alter protein structure/function; Observed in an individual with melanoma (Li et al., 2020); This variant is associated with the following publications: (PMID: 31567591)
This sequence change replaces alanine, which is neutral and non-polar, with glycine, which is neutral and non-polar, at codon 1780 of the POLE protein (p.Ala1780Gly). This variant is present in population databases (no rsID available, gnomAD 0.006%). This missense change has been observed in individual(s) with multiple melanomas (PMID: 31567591). ClinVar contains an entry for this variant (Variation ID: 405792). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt POLE protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
not specified Uncertain:1
Hereditary cancer-predisposing syndrome Uncertain:1
The p.A1780G variant (also known as c.5339C>G), located in coding exon 39 of the POLE gene, results from a C to G substitution at nucleotide position 5339. The alanine at codon 1780 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
