This variant is present in population databases (rs201950040, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with POLE-related conditions. The arginine at codon 1580 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species.
