This variant is present in population databases (rs147088333, gnomAD 0.03%). This variant has not been reported in the literature in individuals affected with POLE-related conditions. ClinVar contains an entry for this variant (Variation ID: 405675). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt POLE protein function with a negative predictive value of 80%. This variant has been reported in an individual with Ewing sarcoma (PMID: 33332384). To our knowledge, this variant has not been reported in the literature in individuals with POLE-related disease. The p.G1352A variant (also known as c.4055G>C), located in coding exon 32 of the POLE gene, results from a G to C substitution at nucleotide position 4055. The glycine at codon 1352 is replaced by alanine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
