This sequence change replaces threonine, which is neutral and polar, with serine, which is neutral and polar, at codon 1146 of the POLE protein (p.Thr1146Ser). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with POLE-related conditions. ClinVar contains an entry for this variant (Variation ID: 540802). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. Hereditary cancer-predisposing syndrome Uncertain:1
The p.T1146S variant (also known as c.3437C>G), located in coding exon 28 of the POLE gene, results from a C to G substitution at nucleotide position 3437. The threonine at codon 1146 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
